Exendin-4 protects retinal pigment epithelial cells against oxidative stress in vitro via PI3K/Akt-STAT3 activation

چکیده
The glucagon-like peptide 1 (GLP-1) analogue Exendin-4 (Ex-4) could perform anti-oxidative effects against oxidative damage both in vitro and in vivo. However, little was known about the direct protection of Exendin-4 on RPE cells against oxidative stress and its underlying mechanism. The present study examined the protective effects of Exendin-4 on RPE cells against H2O2 treatment in vitro and retina I/R injury in vivo. The results indicated that Ex-4 exerted protective effects under H2O2 condition on RPE cells and attenuated the oxidative stress-induced damage, as evidenced by the results of cellular viability, cytotoxicity, apoptosis and western blotting. However, the protective effects of Ex-4 were mainly abrogated by p-Akt inhibitor or p-STAT3 inhibitor. In conclusion, the Ex-4 was able to protect RPE cells from H2O2 induced oxidative damage in vitro and attenuate retinal injury from I/R in vivo, which was closely related to PI3k/Akt and p-STAT3 signaling.

Squamosamide Derivative FLZ Protects Retinal Pigment Epithelium Cells from Oxidative Stress through Activation of Epidermal Growth Factor Receptor (EGFR)-AKT Signaling
Reactive oxygen species (ROS)-mediated retinal pigment epithelium (RPE) cell apoptosis is attributed to age-related macular degeneration (AMD) pathogenesis. FLZ, a novel synthetic squamosamide derivative from a Chinese herb, Annona glabra, has displayed significant cyto-protective activity. In the current study, we explored the pro-survival effect of FLZ in oxidative stressed-RPE cells and stud...

BACKGROUND We evaluated the protective effect of ALS-L1023, an extract of Melissa officinalis L. (Labiatae; lemon balm) against oxidative stress-induced apoptosis in human retinal pigment epithelial cells (ARPE-19 cells). METHODS ARPE-19 cells were incubated with ALS-L1023 for 24 h and then treated with hydrogen peroxide (H2O2). Oxidative stress-induced apoptosis and intracellular generation ...
متن کاملAstaxanthin protects ARPE-19 cells from oxidative stress via upregulation of Nrf2-regulated phase II enzymes through activation of PI3K/Akt
PURPOSE Oxidative stress on retinal pigment epithelial (RPE) cells is thought to play a crucial role in the development and progression of age-related macular degeneration. Astaxanthin (AST) is a carotenoid that shows significant antioxidant properties. This study was designed to investigate the protective effect of AST on ARPE-19 cells against oxidative stress and the possible underlying mecha...
متن کاملCannabinoid receptor 1 blockade protects human retinal pigment epithelial cells from oxidative injury
BACKGROUND Because oxidative stress is assumed to be a key mechanism in the pathological process of age-related macular degeneration (AMD), increasing numbers of studies have focused on discovering new pathways and treatments for reducing oxidative damage. Our work investigates the potential role of the cannabinoid receptor 1 (CB1) in oxidative stress of primary human retinal pigment epithelial...
متن کاملSC79 protects retinal pigment epithelium cells from UV radiation via activating Akt-Nrf2 signaling
Excessive Ultra-violet (UV) radiation causes oxidative damages and apoptosis in retinal pigment epithelium (RPE) cells. Here we tested the potential activity of SC79, a novel small molecule activator of Akt, against the process. We showed that SC79 activated Akt in primary and established (ARPE-19 line) RPE cells. It protected RPE cells from UV damages possibly via inhibiting cell apoptosis. Ak...
